Libraries for DNA sequencing were made by Qingyi (John) Zhang in the J. Eisen lab. More detail coming soon.
454 sequencing was done on a Roche GS Junior forE. fetida. Libraries were constructed according to standard protocols.
Illumina sequencing was done on both a MiSeq and Hiseq system. Libraries were made using a transposes driven strategy. Each of four samples (two of which were Eisenia) were given unique barcodes during the library construction and samples were pooled. Barcode 72 (#447) is fromWorm #1 (thought to be Eisenia fetida).Barcode 70 (#437) is from Worm #2 (thought to be Eisenia hortensis).A single MiSeq run was carried out on the pooled samples at the UC Davis Genome Center Core Facility.In addition a single lane of HiSeq was carried out also using the pooled samples. This run was done as a part of a training run in a new facility and did not work perfectly.Unfortunately the barcode/index sequencing was not done. However, this created an opportunity to test metagenomic assembly and binning methods.
